A SERS platform for determination of Dydrogesterone using CQD-gold nanoparticle conjugate

Dydrogesterone is a selective agonist of the progesterone receptor, with greater oral bioavailability and is commonly used for regulating certain life-sustaining processes in living organisms [1]. Recently, sensors based on carbon quantum dots are used for biological measurements [2]. In this work a surface enhanced Raman scattering (SERS) platform was developed for determination of Dydrogestrone. Carbon quantum dots were synthesized by hydrothermal method and characterized using HRTEM, DLS, UV-Vis spectrophotometry and spectrofluorimetry. Gold nanoparticles were synthesized by reduction of chloroauric acid and also characterized. Different types of carbon quantum dots were synthesized and covalently conjugated to gold nanoparticles. The interaction mechanism of Dydrogestrone with different type of carbon nano dots and different gold nanostructures were studied. A novel conjugate of carbon quantum dots and gold nanoparticles was developed as a SERS platform for the determination of Dydrogesterone. The fluorescence of carbon quantum dots quenched in CQD-GNP conjugate and 2-fold increase in the Raman intensities was observed. The lower limit of quantitation of 10 µg mL-1 (RSD:22.9%) with a linear dynamic range of 10–50 µg mL-1 were obtained for the determination of Dydrogestrone.
