Gold Nanoparticle and Carbon Quantum Dot-Based Nanobiosensor for Imaging and Quantification of Cellular Glutathione Levels

Carbon quantum dots (CQDs) are fluorescent carbon nanoparticles that, due to their low toxicity, strong fluorescence emission, ease of synthesis, low cost, environmental compatibility, and wide range of applications, have emerged as a promising alternative in bioimaging, drug delivery, and chemical sensors [1]. Glutathione (Scheme 1) is an essential and natural antioxidant consisting of three peptides that plays a crucial role in detoxification, neutralizing reactive oxygen species, enhancing the function of other antioxidants, and can be used as an important cancer biomarker [2]. Herein, an off-on fluorescent nanobiosensor was developed for the detection of cellular glutathione using a CQD and gold nanoparticle (GNP) conjugate. The sulfur-doped CQDs were synthesized hydrothermally and characterized using HRTEM, DLS, spectrophotometry, and spectrofluorimetry. The sulfur-modified GNPs were synthesized by sulfide reduction of chloroauric acid and also characterized. The GNPs were conjugated to CQDs using a disulfide bond and quenched their fluorescence by Förster resonance energy transfer (FRET). The imaging and quantification mechanism is based on the fluorescence recovery of the CQDs after GNP-CQD cleavage by the disulfide bond reduction ability of GSH. The nanobiosensor showed high selectivity for glutathione and was applied for the determination of glutathione with a linear dynamic range of 3.0 - 20.0 mM and a detection limit of 1.0 mM. It was also applied for imaging of cellular glutathione levels.
