Immobilization of Lysozyme in ZIF-8 for Enhancing Reusability and Activity of enzyme

A subclass of Metal-organic framework known as Zeolitic Imidazolate Frameworks (ZIFs), which are made of Zn ions as metal nodes and 2-methylimidazole as an organic ligand, typically consists of huge cavities connected by small windows. Due to gentle synthesis conditions, enormous specific surface area, and great water stability, ZIF-8 has been used widely as a template for enzymes immobilization1. The aim of the present study has been to immobilize an enzyme using a metal-organic framework in order to enhance the enzymatic reaction. Lysozyme, as a very common and well-characterized enzyme, was used to immobilize in ZIF-8. As shown in Scheme I, immobilization was done by de-novo encapsulation method2. The Fourier Transform Infrared spectra indicated that the lysozyme was well immobilized in ZIF-8. Scanning Electron Microscopy and X-ray diffraction analysis were also performed, indicating that the sodalite topology for immobilized lysozyme was similar to simulated ZIF-8 and the presence of the enzyme in the structure did not change the ZIF structure. To examine the activity and reusability of the encapsulated enzyme, immobilized lysozyme was used in some C-C coupling reactions. The results indicated that the yield of the reaction and its reusability were significantly enhanced as compared to nonencapsulated enzymes.
