Synthesis and multifaceted functionalization silica nano hollow sphere for controlled drug delivery of zidovudine

Zidovudine is an antiretroviral drug that can cause adverse effects when administered systemically. In this study, we synthesized and functionalized hollow silica nanoparticles (HSNPs) as drug carriers for controlled and targeted delivery of zidovudine to HIV-infected cells. HSNPs were prepared by using a microwave-based sol-gel method with fructose as a sacrificial template. The resulting HSNPs had uniform size, shape, and porosity, and high water retention capability. Zidovudine was loaded into HSNPs by using a solvent evaporation method, achieving a high loading efficiency and stability. HSNPs were further modified with polyethylene glycol (PEG) to enhance their biocompatibility and stealth properties, and with aptamers to improve their specificity and affinity to HIV-infected cells. The in vitro release of zidovudine from HSNPs showed a sustained and pH-dependent profile, indicating the potential of HSNPs for controlled drug delivery. The cellular uptake and cytotoxicity of HSNPs were evaluated using human peripheral blood mononuclear cells (PBMCs) and CEM-SS cells, a T-lymphoblastoid cell line infected with HIV-1. The results demonstrated that HSNPs were efficiently internalized by CEM-SS cells, and exhibited higher antiviral activity and lower cytotoxicity than free zidovudine. These findings suggest that HSNPs are promising nanocarriers for controlled and targeted delivery of zidovudine to HIV-infected cells, which could improve the therapeutic outcome and reduce the side effects of zidovudine treatment.
