A new one-pot synthesis of 5,5-Dimethylhydantoin

The hydantoin scaffold is a versatile structure that has shown potential in various medicinal applications, including as androgen receptor modulators, anticonvulsants, [1] antidiabetic agents, and anticancer agents. This makes it an important target for synthetic chemists looking to develop new drugs. [2, 3] In this study, we focused on developing an efficient method for the synthesis of 5,5-disubstituted hydantoin. We used a reaction between a ketone compound and ammonium carbonate, along with potassium cyanide as a catalyst-free system. By optimizing the reaction conditions, we were able to achieve good yields of the desired hydantoin derivatives. The absence of a catalyst simplifies the synthetic procedure and reduces costs associated with catalysts. This research contributes to the further development of synthetic protocols for hydantoin derivatives, which potentially leads to the production of drugs on an industrial scale.
