Ultrasonicated Green Synthesis of Isoxazolones using Sodium Cyclamate

Nitrogen and oxygen-containing heterocycles, including isoxazoles, serve as key structural units that are found in pharmaceuticals, natural products, some marketed drugs, agrochemicals, functional materials, and organic dyes [1]. Isoxazolone heterocycles are among the most attractive isoxazoles that have a wide range of biological properties, including anticancer, anti-inflammatory, antimycobacterial, antibacterial, antiviral, antitubercular, anti-Alzheimer's, antioxidant, insecticide, and anti-nociceptive. These heterocycles will provide fruitful leads for future open innovation drug discovery [2-4]. In this research investigation, the three-component cyclocondensation route was applied to synthesizing isoxazolones (Scheme 1). Isoxazolone compounds were synthesized by a cyclization of various aldehydes with hydroxylamine and β-oxo esters using ultrasound waves. Optimization studies revealed that 5 mol% of sodium cyclamate in water at room temperature is sufficient to perform the experiment. In these experiments, water was used as a green solvent and ultrasonication was applied as the non-conventional source of green energy. The structures of the synthesized heterocycles were identified via spectroscopic methods (FT-IR and NMR spectroscopies) and elemental analysis.
